Psilocybin Adventures

I’ve never done agar before. I’ve always done liquid culture.

I can provide a recipe if interested.

Absolutely. I also purchased a liquid culture premix powder. What’s the best way to multiply. I have no more LC left. But can’t I use mycelium from one of the grain jars. I think I seen where you inject the grain jar that is colonized with the multiplier and then suvk it back up in your syringe. Then put back into the multiplier.

So you all have peaked my curiosity on all this, so I got a mono tub kit, a sab and just received spores from the spore source. Don’t know if that’s a good company to go thru but giving them a try, I’ve spent the last couple weeks reading through this thread(8k+) and read thru @Caligurl thread as well and also going thru shroomery website now. I’ve squeezed a lot of info into my thimble sized brain and with any luck maybe retained some :crossed_fingers:. Planing on using agar , seems simple enough, biggest hurdle being cleanliness. I’ll for sure keep y’all posted as I go down this rabbit hole. Stay lit everyone :sign_of_the_horns:

Welcome.

I just realized that post is 4 years old. I’ve been doing this for at least 5 years.

I gots some pre sterilized agar petri dishes coming from north spore, should be getting 20 on or around Saturday so gonna be slowly getting my crap together and set up hopefully over the weekend and start working on my sterile skills I’ll post everything here as I go thru this. I’m very open to suggestions and any input in general, this is a whole new rabbit hole for me. And as always stay lit y’all :smiling_face_with_sunglasses::fire::melting_face:

Do you do agar at all or just spores to grain? I’m gonna play some this afternoon on doing a few drops of spores on a couple plates and see how it goes. Gotta practice using the sab anyway and figure out best methods as well, some trial and error you might say :sign_of_the_horns:

I inoculate popcorn in pint jars with ~3ml of spore solution.

Never done agar, but I usually maintain liquid culture jars in my Martha tent.

Hopefully I can get a handle on the agar, everything I’m reading definitely recommends learning agar as you can make sure you start with no contamination and can do everything else as well, LC, agar 2G, ect… . I’ll post some stuff later when I start playing :sign_of_the_horns::melting_face:

Similar to @MidwestGuy, I inoculate grain with liquid cultures and then spawn to bulk.

My experience with colonization times:

Spores to grain (in a pint jar): 6 weeks to full colonization

Liquid culture to grain (pint jar): 4 weeks to full colonization.

I have one 10ml spore syringe of golden teacher left and need to use it for LC.

LC recipe: RO or distilled with 3/4 teaspoon of Karo syrup for mycelium nutrients. Be careful how much Karo syrup you use. If the water/sugar ratio is 5% by volume, then it is perfect. Too much or too little and mycelium will not grow.

You can turn a single syringe of commercial spore solution into dozens of flushes by maintaining LC jars.

I failed to mention that this recipe is for a half-pint jar.

Here’s my s.a.b. , played a little with petri dishes in the sab, trying to get more familiar with steril tek as in slow movements and not reaching over sterile objects and such. Gonna take some practice, so I did dribble some stargazer spores onto 3 petri dishes and sealed em up , I did try using Saran wrap but didn’t go so well, I guess you can cut an 1" 1/2 section and wrap using that but wasn’t working for me so went with parafilm and worked much better I also inoculated the two grain bags in my kit, so now we’ll wait and see what comes out I have 5 more different syringe’s in the fridge yet so definitely more to come :sign_of_the_horns::smiling_face_with_sunglasses::melting_face:

@MidwestGuy
I’ve transferred to bulk. Noticed a lot of condensation on the lids. (Same as yours) I’m going to take a pic tonight. Was curious if you also have condensation that build up on your tote lids while it’s colonizing.

I always get lid condensation. I try to open the tub and wipe it off if it is heavy, but only if absolutely necessary. Don’t like opening my tubs and introducing contaminants.

Keep in mind that substrate that is too wet will not colonize.
