Inducing Polyploidy in Cannabis Using Colchicine Extracted from Saffron (Crocus sativus)

Here’s the best top from one of the clones from my Sleepy Joe OG poly.

It weighed maybe 3g the stem reminded me of celery. Here’s a normal bud from the same plant

I ended up letting it go about a week too long. I was out of town the week I should have harvested it. Too many amber trichomes. It was strong and straight up couch lock weed, so I was told. I was growing for my wife and MIL I wasn’t smoking back then. It also got dried way too fast and to hot. Made it taste like crap. Luckily I grew a Banana Kush that went a few weeks into November before it was ready.

:joy: first i thought it’s two stem beside each other, cool!
i didn’t knew there is a black widow strain, look at that color, fascinating.

You didn’t smoke yourself, but you still went through the trouble of growing it for your wife and MIL (I don’t know who or what that is—is it a name?). You have my respect!

Man, that Black Widow is hypnotizing me.

How was it’s potency? Is it right that some people say poly ones are weak?

I grew Black Widow again in 2021 and 2023 It was some of the best weed Ive grown and definitely the prettiest weed ive ever grown. MIL stands for Mother in law. I started smoking weed again in 2020 after my pain management doctor recommended it to help when my pain meds weren’t doing enough. I refused to go on opiates and the meds I was taking had a limit on how much can be prescribed. It was either weed or opiates. I chose weed.
Heres a picture from 2021

This one is from 2023

No doubt, it’s beautiful. Oh, so that’s what it means—I’m sorry to hear about having pain and that… I truly hope things get better for you every day. Having worked in a pharmacy, I completely understand your situation. Choosing weed over opiates is definitely a wise decision.

The potency is probably higher in the polyploids, at least the ones I’ve grown. I tagged clones from a regular Sleepy Joe with one colour of collar, and clones from the poly with a different colour. When I dried and jarred them, I labelled the jars as “poly”. The poly jars have a much nicer flavour profile, and maybe a bit more potent. SJ is supposed to be 33% THC, so hard to tell strength difference (did I just get hit with an 8 pound hammer or a 10 pounder?).

Can’t wait for my next polyploid!

After 4 days, the sprayed seeds look a few steps behind. At first, I was sad, thinking I did something wrong, but then I realized—this might actually mean it’s working! Colchicine could be causing polyploidy (double chromosomes), making cells larger and slower to regenerate. Bigger cells take more time to grow, so the delay might be a good sign. I’m keeping my hopes up and watching closely for changes! :seedling:

Edit:
I forgot to add that the difference between treated and untreated seeds is not that big, I just wanted to show the average of each.
The treated ones do have bigger and better sprout, but only 3 out of 10. While the untreated ones have 6 out of 10 large and good sprout.

I’m starting to think I actually managed to extract colchicine, but I still need more information to be sure.
Right now, I see two potential problems:

  1. The dosage: I might be using too much
  2. Timing: I started the treatment too early.

To figure this out, I planted three groups of untreated plants:

  1. Control: No colchicine at all.
  2. Single Dose: One small dose of colchicine.
  3. Continuous: Regular doses over time.

My goal is to figure out if I’m actually mutating these plants (inducing polyploidy) or just poisoning them. The differences in their growth could have a lot of explanations, so I’m keeping a close eye on their development.

An update of this test subject:

The new main branch naturally split into two sub-branches and began to grow. After I topped them, it appears that another branch is starting to grow in the middle of those two branches.

Do you think this is normal or my C.C.E did that?

If C.C.E did that the next and final test will be on cannabis plant. ( in the middle of veg stage )

So here’s my cannabis experiment from the beginning to now.
I diluted my solution about 50% and sprayed it on the plant every other day.

Yo!
Hope you all are doing good

So i have news, one day i was watering my chosen test subject with CCE and accidentally it got over watered… like a lot!
So i poured the extra liquid in my test subject pot to the one beside it, i thought that test sub #1 will die now because of my #uck up so lets start with another.
I found out that I’m using really little cups so i repotted them and i found out that test sub #1 that i over watered is having root rot due to bad drainage holes.

The interesting part is that the plant which i poured the extra CCE in it, actually mutated to a triploid so i call it test sub #2 now:

It also was stunned for a while, like test sub #1 which makes me think that perhaps if the first one had better drainage holes and haven’t got root rot maybe it would mutated too! Or im still giving them a killer dose.
The first test subject is recovering so i will have my eyes on it, new leafs should show any signs of mutation.
I’m also considering that the date of my CCE has expired, i havent been keeping it in a dark and cold location so it is logical that colchicine would react with something in there and my CCE is losing or has already lost its potency.
